Résumé
L’imagerie dans le proche infrarouge offre des avantages importants en optique biomédicale grâce à différentes méthodes complémentaires permettant d’obtenir de l’information sur l’état des tissus biologiques de manière non invasive. Ce mémoire présente la conception et la validation d’une plateforme d’imagerie multimodale combinant la fluorescence endogène, la fluorescence exogène et une méthode d’imagerie Raman multispectrale. Il met en valeur le potentiel de recherche et la combinaison de diverses approches d’imagerie dans le proche infrarouge. Il applique ces possibilités à l’imagerie grand champ de tissus tumoraux cérébraux et intègre l’utilisation de marqueurs moléculaires tels que les nanotubes de carbone et leur combinaison avec des fluorophores exogènes. Il introduit aussi une méthode de correction du signal selon les propriétés optiques des tissus et une méthode multispectrale isolant le signal Raman à l’aide de seulement trois filtres. L’application de ces technologies pour l’identification de tissus cancéreux ou de marqueurs ciblant les tumeurs constitue son objectif principal. La première modalité du système vise à imager la présence de nanotubes de carbone capables de cibler les tumeurs en s’y accumulant. Cette partie cherche à identifier la fluorescence émise par des fluorophores liés aux nanotubes ou à les détecter directement à partir de leur signal Raman, notamment dans les bandes D (1350 cm⁻¹) et G (1600 cm⁻¹). Le système est conçu pour minimiser les pertes de transmission du signal surtout à la longueur d’onde du Raman. Il cible un pic Raman à l’aide de trois filtres permettant d’observer l’intensité du signal tout en soustrayant l’autofluorescence. La seconde modalité utilise la fluorescence endogène des tissus biologiques pour caractériser les tissus tumoraux du cerveau. Cette étude s’appuie sur des spectres Raman in vivo (430 mesures, 26 patients), avec une valeur prédictive positive de 85–88 % pour la détection tumorale et une valeur prédictive négative de 96 % pour le tissu cérébral normal. Le montage grand champ a été adapté pour l’acquisition d’images de fluorescence et de réflectance sur des échantillons ex vivo. Les résultats ont été obtenus sur 14 échantillons provenant de 5 patients lors de leur opération du cancer du cerveau. Le système élimine les réflexions spéculaires et applique une méthode de correction pour tenir compte des variations optiques et géométrique. Les résultats démontrent un contraste intrinsèque robuste, avec des différences significatives (p < 0,05), ouvrant la voie au développement d’outils optiques pour l’assistance à la visualisation des tissus tumoraux.
Abstract
Near-infrared imaging offers significant advantages in biomedical optics through various complementary methods that enable the collection of information on the state of biological tissues in a non-invasive manner. This thesis presents the design and validation of a multimodal imaging platform combining endogenous fluorescence, exogenous fluorescence, and a multispectral Raman imaging method. It highlights the research potential and the combination of different near-infrared imaging approaches. It applies these capabilities to wide-field imaging of brain tumor tissues and integrates the use of molecular markers such as carbon nanotubes and their combination with exogenous fluorophores. It also introduces a signal-correction method based on the optical properties of tissues and a multispectral approach isolating the Raman signal using only three filters. The application of these technologies for identifying cancerous tissues or tumor-targeting markers constitutes its main objective. The first modality of the system aims to image the presence of carbon nanotubes capable of targeting cancerous tumors by accumulating within them. This part of the project seeks to identify the fluorescence emitted by fluorophores linked with the nanotubes or to directly detect them through their Raman signal, particularly in the D (1350 cm⁻¹) and G (1600 cm⁻¹) bands. The system is designed to minimize overall signal losses especially at the Raman wavelength. It targets a Raman peak using three filters that allow both observation of signal intensity and subtraction of the autofluorescence component. The second modality uses the endogenous fluorescence of biological tissues to characterize brain tumor tissues. This study is based on fluorescence signatures observed in in vivo Raman spectra from 430 measurements on 26 patients, yielding a positive predictive value (PPV) of 85–88% for tumor detection and a negative predictive value (NPV) of 96% for normal brain tissue. The wide-field setup was adapted for the acquisition of fluorescence and reflectance images on ex vivo samples. The results were obtained on 14 samples from 5 patients during their brain cancer surgery. The system eliminates specular reflections and applies a correction method to account for spatial variations in illumination, surface geometry, and tissue absorption and scattering properties. Overall, the results demonstrate a robust intrinsic contrast, with statistically significant differences (p < 0.05) across raw and corrected data as well as paired and grouped analyses, providing a foundation for the development of optical tools aimed at assisting tumor tissue visualization.
