Préparation de films à base de chitosane par voie fondue

Résumé

Depuis plusieurs années, le besoin d'utiliser des polymères d'origine naturelle dans l'industrie des plastiques se fait de plus en plus pressant. Le chitosane, qui est un polysaccharide naturel, a en plus des propriétés antibactériennes qui peuvent être intéressantes dans de nombreux domaines comme l'industrie de l'emballage alimentaire. L'inconvénient dans l'utilisation du chitosane est qu'il ne présente pas de point de fusion avant sa dégradation, il n'est donc pas adapté aux méthodes traditionnelles de transformation des polymères. L'objectif de ce travail a été de développer une nouvelle technique pour préparer des films antibactériens à base de chitosane par voie fondue. La plastification du chitosane a été choisie en assumant que cette plastification, qui est déjà réalisée en solution, est possible à l'état fondu et qu'elle permettra d'obtenir une bonne répartition du chitosane à la surface d'un film polymérique. La plastification du chitosane s'effectue à l'aide d'un traitement thermomécanique et en présence d'un solvant et d'un plastifiant. Trois plastifiants ont été sélectionnés dans la famille des polyols : le glycérol, le xylitol et le sorbitol par ordre croissant de poids moléculaire. Une solution d'acide acétique à 3 % en volume a été choisie comme solvant. Plusieurs rapports chitosane/polyol ont été utilisés dans la formulation du chitosane plastifié. Le chitosane a été plastifié dans un mélangeur interne à 80 °C, à une vitesse de 100 rpm pendant 15 min. Pour la première fois, la morphologie du chitosane plastifié a été observée au MEB. Elle consiste en une phase cristalline, composée de feuillets successifs, incorporée dans une phase amorphe. Des réactions entre l'acide acétique et le chitosane et entre les polyols et le chitosane ont été observées grâce à la FTIR. Il a été montré que plus le poids moléculaire du polyol était bas, moins la cristallinité du chitosane plastifié était élevée. Le chitosane plastifié avec le glycérol a démontré la phase amorphe la plus importante, ses différentes propriétés mesurées étant alors les plus faibles. En admettant que la mise en oeuvre du chitosane plastifié avec le glycérol serait facilitée grâce à l'importance de sa phase amorphe, le glycérol a été préféré aux deux autres polyols dans le reste de l'étude. La quantité de polyol a aussi son impact sur les propriétés, plus celle-ci augmente plus la phase amorphe est importante donc plus les propriétés sont faibles mais sa mise en oeuvre aisée. Pour comprendre l'effet de chaque composant, différents pourcentages volumiques d'acide acétique en solution, plusieurs ratios chitosane/solution d'acide acétique et plusieurs ratios chitosane/glycérol ont été utilisés pour plastifier le chitosane. Le chitosane plastifié a été mélangé vii avec un polyéthylène métallocène (mPE) dans le mélangeur interne. La morphologie des ces mélanges a été observée au MEB et corrélée avec leurs propriétés rhéologiques grâce au modèle de Palierne. La présence de l'acide acétique engendre une déstructuration du chitosane par une détérioration des liaisons hydrogènes entre les plans du chitosane. Sous cette action, les plans se sont séparés et un gonflement du chitosane dans la matrice a été observé. La partie aqueuse permet le mouillage de la surface du chitosane. Le glycérol s'est glissé entre les plans écartés du chitosane, le gonflement s'est poursuivi jusqu'à la rupture de la structure et la réduction de la taille des domaines de chitosane. Lorsque la quantité de glycérol dans la formulation est augmentée, la dispersion du chitosane dans la matrice de polyéthylène est améliorée. Les mélanges chitosane plastifié/mPE ont montré de bonnes propriétés antibactériennes avec une réduction de 2 log (Colony Forming Unit/mL) de la population totale de bactéries Escherichia coli par rapport au mPE pur. Les propriétés mécaniques ont été améliorées par la compatibilisation avec l'éthylène-acétate de vinyle (EVA) qui s'est positionné à l'interface entre le mPE et le chitosane. Afin d'améliorer la plastification du chitosane, un changement dans sa mise en forme a été réalisé. La plastification du chitosane et son mélange avec un polymère fondu a été réalisée en une seule passe dans une extrudeuse bivis. Le chitosane a été plastifié en amont de la bivis, le mPE étant quant à lui fondu dans une extrudeuse monovis connectée au milieu de la bivis. Le chitosane plastifié et le mPE fondu ont alors été mélangés en aval de l'extrudeuse bivis. Des films ont été par la suite extrudés à 2, 5 ou 10 % en poids de chitosane, avec ou sans l'EVA pour la compatibilisation. La plastification a grandement été améliorée par le changement de mise en forme. Les images des films réalisées au MEB, ont montré une réduction par 5 de la taille des domaines de chitosane par rapport aux mélanges produits avec le mélangeur interne. Cet aspect s'est répercuté sur l'aspect visuel qui a été fortement amélioré. La dégradation thermique des films a été accélérée par l'ajout du chitosane plastifié et les propriétés mécaniques ont été affectées négativement par l'ajout du chitosane. La compatibilisation avec l'EVA a malgré tout aidé à maintenir de bonnes propriétés mécaniques pour les plus faibles quantités de chitosane. De plus, l'ajout du chitosane plastifié n'a eu aucun impact sur les propriétés barrières à l'oxygène des films. Finalement, aucune activité antibactérienne n'a été mesurée en présence des films. La présence du chitosane à la surface du film n'a pas été observée et cela serait directement lié à viii l'absence de ces propriétés antibactériennes. En raison de l'efficacité du mélange dans l'extrudeuse, l'encapsulation du chitosane par l'EVA devait être complète.

Abstract

For many years, the plastics industry has considered the use of polymers of natural origin. Certain specific characteristics of these natural polymers have also been investigated to increase the added value of the final plastic product. Chitosan can be a good polymer candidate since it is a natural polysaccharide and it exhibits interesting properties such as antibacterial properties that can apply to several domains such as food packaging industry. The inability of chitosan to melt before its thermal degradation makes it unsuitable for traditional polymer processing methods. The objective of this work is to develop a new technique to prepare chitosan-based anti-bacterial films through a molten route. This approach was selected according to the hypothesis that the plasticization of chitosan – already performed by the solvent casting method – is possible in the molten state and would provide a good chitosan distribution at the surface of a polymeric film. In this work chitosan plasticization is achieved by a thermo-mechanical treatment in the presence of an acidic liquid phase and a plasticizer. Three plasticizers were selected from the polyol family: glycerol, xylitol and sorbitol, presented from low to high molecular weight. A 3 % v/v solution of acetic acid was chosen as the solvent. Plasticized chitosan was prepared with various chitosan/polyol ratios and was processed in an internal mixer at 80 °C for 15 min using a rotation speed of 100 rpm. The samples were then hot-pressed to produce sheets of 1 mm thickness. SEM micrographs demonstrated a crystalline phase, composed of a layered structure, embedded in an amorphous phase. The interactions between chitosan and the acetic acid solution and between chitosan and polyols were investigated by FTIR analysis. It was observed that the crystallinity of the plasticized chitosan increased with polyol molecular weight. Plasticized chitosan using glycerol had the highest content of the amorphous phase and the lowest values of the investigated properties. Moreover, as the polyol content increased the crystallinity decreased. Due to the dominant amorphous phase content with glycerol, it was assumed that chitosan processing would be facilitated. As a result, glycerol was favored over the other two polyols for the continuation of the study. In order to understand the role of each component in the plasticization mechanism, the following parameters have been examined: acetic acid content, chitosan/acetic acid solution ratio and chitosan/glycerol ratio. These various plasticized chitosans were melt-blended with a metallocene polyethylene (mPE) in an internal mixer. The blends morphology was examined by SEM and x was related to the rheological properties using the Palierne model. Acetic acid allowed chitosan destructuration by breaking down the hydrogen bonds between its structural sheets. The distance between two planes increased and swelling of chitosan embedded in the mPE matrix was observed. The acidic aqueous phase of the formulation was essential to thoroughly wet the chitosan powder. As glycerol penetrated between the spaced sheets, the swelling of chitosan domains continued until structural rupture occurred, which led to a reduction of chitosan domains size. SEM images confirmed that an increase in the glycerol content led to a better chitosan dispersion in the mPE matrix. Plasticized chitosan/mPE blends demonstrated good antibacterial properties with a reduction by an order of 2 log(Colony Forming Unit (CFU)/mL) in the number of bacteria Escherichia coli with respect to neat mPE. Ethylene-vinyl acetate (EVA) was also added in the blend to promote compatibilization between chitosan and mPE. It was found that EVA was located around chitosan domains, according to SEM imaging. This compatibilization step improved the mechanical properties of the blends. In order to improve further chitosan plasticization, the processing technique was modified. Chitosan plasticization and its blend with a molten polymer was achieved in a single-pass extrusion process. Chitosan was plasticized in the first zone of a twin-screw extruder. mPE was melted in a single-screw extruder connected to the center of the main twin-screw extruder and was injected inside the latter into the second zone. Then, plasticized chitosan and mPE were blended together. Dilutions were performed in single screw extrusion to produce different film formulations with 2, 5 and 10 wt% of chitosan with and without EVA as the compatibilizer. It was shown that such modification of the processing technique greatly improved plasticization. SEM images demonstrated that the size of the chitosan domains were reduced by a factor of 5 with respect to the blends prepared by internal mixing. Therefore optical properties were also improved. However the thermal degradation of the films was accelerated in the presence of plasticized chitosan. Mechanical properties were also negatively affected, although compatibilisation with EVA allowed maintaining them to an acceptable level for the lowest chitosan contents. Oxygen permeability of the films remained constant after the addition of chitosan, while water vapor permeability increased. Finally, no antibacterial properties were revealed for the newly produced films. It is speculated that the absence of antibacterial characteristics is related to the fact that no chitosan was detected at the film surface, probably due to chitosan encapsulation by EVA as a result of the high efficiency of mixing.
